Abstract

The invention relates to a process for the preparation of a bi-functionalized conjugate, wherein the conjugate is a peptide-oligopeptide or a peptide-oligonucleotide conjugate (POC), the process comprising the steps of: (i) providing a peptide according to structure (1), or a mixture of more than one peptide according to structure (1), wherein: G is a peptide comprising 1–50 amino acids; and AA1 and AA2 are independently selected from the group of amino acid side chains; AA3 is a tyrosine, tryptophan, arginine or lysine side chain; (ii) modifying the C-terminus of the peptide via click chemistry; and (iii) modifying the N-terminus of the peptide via click chemistry. The C-terminus and the N-terminus of the peptide are modified in such a way that the amino acid sequence of for example the POC peptide moiety may be determined, preferably by nanopore sequencing. The invention also relates to the bi-functionalized peptide-oligopeptide conjugates and POCs themselves.
